I don't think BizTalk was ever "formally" supported in a virtual environment - but given that is now out of main-stream support this is kinda irrelevant. It certainly works just fine in a virtualised environment and we have many running virtual.
I believe is licensed per physical socket. It doesn't matter how many cores or virtual processors allocated to a VM.
That said I believe if you tie down a VM to a single socket you don't have to license multiple sockets providing you can guarantee it doesn't shift between processors.
